In healthy study subjects, administration of a single 200 mg dose of abemaciclib during 600 mg daily dosing of rifampin, a potent CYP450 3A4 inducer, decreased the relative potency-adjusted unbound systemic exposure (AUC) of abemaciclib plus its active metabolites by 67% relative to abemaciclib administered alone. Reduced efficacy of abemaciclib may occur.

MANAGEMENT: Concomitant use of abemaciclib with potent CYP450 3A4 inducers should generally be avoided.
