Relevance: 20.07.2022 Reviewer: Shkutko P.M., M.D., in

In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. A doctor should be consulted to address the issue of joint drug administration.

Consumer:

CarBAMazepine can significantly reduce the blood levels of abiraterone, which may make the medication less effective in treating your condition. Talk to your doctor if you have any questions or concerns. Your doctor may be able to prescribe alternatives that do not interact, or you may need a dose adjustment or more frequent monitoring to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Professional:

GENERALLY AVOID: Coadministration with potent inducers of CYP450 3A4 may significantly decrease the plasma concentrations of abiraterone, which is partially metabolized by the isoenzyme. In healthy subjects administered a single 1000 mg dose of abiraterone following pretreatment with the potent CYP450 3A4 inducer rifampin (600 mg daily for 6 days), mean abiraterone systemic exposure (AUC) decreased by 55%.

MANAGEMENT: Concomitant use of abiraterone acetate with potent CYP450 3A4 inducers should generally be avoided. Otherwise, an increase in dosing frequency for abiraterone acetate to twice daily during the coadministration period (e.g., from 1000 mg once daily to 1000 mg twice daily) may be considered, although this dosage adjustment has not been studied clinically. The dosage should be adjusted back to the previous level when the CYP450 3A4 inducer is discontinued.
