Somatropin may reduce the blood levels and effects of SUNItinib. Contact your doctor if your symptoms worsen or your condition changes during treatment with these medications. Your doctor may be able to prescribe alternatives that do not interact, or you may need a dose adjustment or more frequent monitoring to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

GENERALLY AVOID: Coadministration with inducers of CYP450 3A4 may decrease the plasma concentrations of sunitinib and its pharmacologically active metabolite, both of which are substrates of the isoenzyme. In healthy volunteers, concurrent administration of a single dose of sunitinib with the potent CYP450 3A4 inducer, rifampin, resulted in a 23% and 46% decrease in the combined (i.e., sunitinib plus its primary active metabolite) peak plasma concentration (Cmax) and area under the concentration-time curve (AUC) values, respectively, compared to administration of sunitinib alone.

MANAGEMENT: Alternative agents with no or minimal CYP450 3A4-inducing activity are recommended in patients treated with sunitinib. A dosage increase for sunitinib to a maximum of 87.5 mg daily should be considered if it must be coadministered with CYP450 3A4 inducers such as barbiturates, dexamethasone, efavirenz, nevirapine, rifamycins, and some antiepileptic agents. Patients should be monitored carefully for toxicity, and the sunitinib dosage reduced if necessary.
