Using amprenavir together with ritonavir may increase the effects of amprenavir. Contact your doctor if you experience increased side effects or if your condition changes. If your doctor does prescribe these medications together, you may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

ADJUST DOSE: The concomitant administration of ritonavir may significantly increase amprenavir AUC and trough concentrations. In healthy subjects, amprenavir AUC increased by 62% to 64% and trough concentrations increased by 319% to 508% with ritonavir 200 mg/day. The mechanism is inhibition of CYP450 3A4 metabolism by ritonavir.

MANAGEMENT: The manufacturer recommends that the adult amprenavir dosage be reduced to 1200 mg once daily with ritonavir 200 mg once daily or 600 mg twice daily with ritonavir 100 mg twice daily.
