Professional:

Albiglutide delays gastric emptying, which may impact the absorption of concomitantly administered oral medications. In pharmacokinetic studies, albiglutide did not affect the absorption of several orally administered medications to any significant extent, except for simvastatin (see below).

Acarbose: Administration of acarbose in patients with intestinal obstruction is contraindicated. Some authorities recommend caution when acarbose is administered concomitantly with albiglutide.

Digoxin: Administration of a single 0.5 mg dose of digoxin with albiglutide at steady state (50 mg once a week for 5 weeks) increased mean digoxin peak plasma concentration (Cmax) by 11% and systemic exposure (AUC) by 9%.

Oral Contraceptives: Administration of ethinyl estradiol-norethindrone (0.035 mg-0.5 mg daily--21 days active/7 days placebo) with albiglutide (50 mg once a week for 4 weeks) increased mean norethindrone Cmax by just 4% and had no change on the AUC. When a different oral contraceptive containing levonorgestrel was given for 28 days with albiglutide, mean levonorgestrel Cmax and AUC increased by 20% and 9%, respectively. No clinically relevant pharmacodynamic effects on luteinizing hormone, follicle-stimulating hormone, or progesterone were observed during coadministration of the oral contraceptives with albiglutide.

Simvastatin: Administration of a single 80 mg dose of simvastatin with albiglutide at steady state (50 mg once a week for 5 weeks) increased mean simvastatin Cmax by 18% but decreased the AUC by 40%. Mean Cmax and AUC of the active metabolite, simvastatin acid, increased by 98% and 36%, respectively. The clinical significance of these changes is unknown.

Warfarin: Administration of a single 25 mg dose of warfarin with albiglutide at steady state (50 mg once a week for 5 weeks) resulted in a 6% decrease and a 2% increase in the mean Cmax and AUC of R(+) warfarin, respectively, while mean Cmax and AUC of S(-) warfarin decreased by 7% and 1%, respectively. Albiglutide did not significantly alter the pharmacodynamic effects of warfarin as measured by the international normalized ratio (INR).
