Consumer:

Using didanosine together with tipranavir may decrease the effects of didanosine. For best results, didanosine and tipranavir should be administered at least 2 hours apart. If your doctor does prescribe these medications together, you may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Professional:

ADJUST DOSING INTERVAL: Coadministration with tipranavir in combination with low-dose ritonavir may decrease the plasma concentrations of didanosine. The mechanism of interaction is unknown. In 10 HIV-positive subjects, tipranavir/ritonavir (250 mg/200 mg twice a day for 21 days) decreased the mean peak plasma concentration (Cmax) and area under the plasma concentration-time curve (AUC) of steady-state didanosine (125 mg or 200 mg twice a day based on weight) by 43% and 33%, respectively, compared to administration of didanosine without tipranavir/ritonavir. In five healthy, HIV-negative volunteers, didanosine (400 mg single dose) reduced the trough plasma concentration (Cmin) of tipranavir (administered as tipranavir/ritonavir 500 mg/100 mg twice a day for 14 days) by 34%. The clinical significance of these changes is unknown.

MANAGEMENT: Antiretroviral response should be monitored closely when tipranavir/ritonavir is used in combination with didanosine. For maximal absorption, didanosine and tipranavir/ritonavir should be administered at least 2 hours apart.
