GENERALLY AVOID: Coadministration with potent inducers of CYP450 3A4 may significantly decrease the plasma concentrations of abemaciclib and its pharmacologically active metabolites, all of which are substrates of the isoenzyme. In healthy study subjects, administration of a single 200 mg dose of abemaciclib during 600 mg daily dosing of rifampin, a potent CYP450 3A4 inducer, decreased the relative potency-adjusted unbound systemic exposure (AUC) of abemaciclib plus its active metabolites by 67% relative to abemaciclib administered alone. Reduced efficacy of abemaciclib may occur.

MANAGEMENT: Concomitant use of abemaciclib with potent CYP450 3A4 inducers should generally be avoided.
