CarBAMazepine may reduce the blood levels and effects of ezogabine. You may need a dose adjustment if you have been taking ezogabine and are starting treatment with carBAMazepine. Similarly, if carBAMazepine is discontinued, your dosage of ezogabine may need to be readjusted. Contact your doctor if your condition changes or you experience loss of seizure control during treatment with these medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

ADJUST DOSE: Coadministration with carbamazepine or phenytoin may decrease the plasma concentrations of ezogabine. The mechanism may involve induction of ezogabine metabolism via uridine 5'-diphosphate-glucuronyltransferases (UGTs). In a phase 2 study, administration of ezogabine 300 to 1200 mg/day in combination with carbamazepine 600 to 2400 mg/day reduced the ezogabine peak plasma concentration (Cmax) and systemic exposure (AUC) by 23% and 31%, respectively, and increased the clearance by 28%. Administration of ezogabine with phenytoin 120 to 600 mg/day reduced the ezogabine Cmax and AUC by 18% and 34%, respectively, and increased the clearance by 33%.

MANAGEMENT: An increase in dosage of ezogabine should be considered when adding phenytoin or carbamazepine. Likewise, the ezogabine dosage may need to be reduced when these agents are discontinued.
