Chloroquine and Methylene blue injection

Professional:Coadministration with methylene blue may modestly decrease the concentrations of chloroquine in whole blood. In 24 healthy volunteers, methylene blue (130 mg orally twice daily for 3 days) decreased the area under the whole blood concentration-time curve (AUC normalized to body weight) of chloroquine (1000 mg orally (males) and 750 mg orally (females) for 2 days, then half the dose on the third day) by 21% and that of the pharmacologically active metabolite desethylchloroquine by 35% compared to placebo. The renal clearance and terminal elimination half-life of chloroquine and the metabolic ratio between chloroquine and desethylchloroquine exposure were not significantly altered. No serious adverse events occurred in the study. This interaction is unlikely to have major relevance on the clinical outcome of chloroquine treatment.
