Coadministration with cimetidine may modestly increase the plasma concentrations of threohydro- and erythrohydrobupropion, two metabolites of bupropion with approximately one-fifth the pharmacologic activity of the parent compound. The mechanism of interaction is unknown. In 24 healthy male volunteers, administration of two 150 mg bupropion sustained-release tablets with a single 800 mg dose of cimetidine following an overnight fast had no effect on the pharmacokinetics of bupropion or hydroxybupropion compared to administration of the bupropion alone. However, there were 32% and 16% increases in the peak plasma concentration (Cmax) and area under the concentration-time curve (AUC), respectively, of threohydro- and erythrohydrobupropion combined. These changes are unlikely to be of clinical significance.
