Consumer:Lumefantrine may increase the blood levels and effects of PARoxetine. You may need a dose adjustment or more frequent monitoring by your doctor to safely use both medications. Contact your doctor if you experience increased side effects or your condition changes. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Professional:MONITOR: Coadministration with lumefantrine may increase the plasma concentrations of drugs that are substrates of the CYP450 2D6 isoenzyme. In vitro, lumefantrine significantly inhibits the activity of CYP450 2D6 at therapeutic plasma concentrations. Adverse effects of drugs that are primarily metabolized by CYP450 2D6 may be increased; however, clinical data have not been reported.
MANAGEMENT: Caution is advised if lumefantrine is used concurrently with medications that undergo metabolism by CYP450 2D6, particularly those with a narrow therapeutic range. Dosage adjustments as well as clinical and laboratory monitoring may be appropriate for some drugs whenever lumefantrine is added to or withdrawn from therapy.
