Professional:Coadministration with probenecid may modestly increase the plasma concentrations of eluxadoline. The proposed mechanism is probenecid inhibition of multidrug resistance-associated protein 2 (MRP2) and organic anion transporter 3 (OAT3), which are involved in the hepatobiliary efflux and proximal tubular uptake of eluxadoline, respectively. In 30 healthy volunteers, administration of a single 100 mg dose of eluxadoline with a single 500 mg dose of probenecid increased eluxadoline peak plasma concentration (Cmax) by 31% and systemic exposure (AUC) by 35% compared to administration of eluxadoline alone. These changes are not considered clinically significant.
