Colchicine and probenecid and Glycerol phenylbutyrate
Determining the interaction of Colchicine and probenecid and Glycerol phenylbutyrate and the possibility of their joint administration.
In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. A doctor should be consulted to address the issue of joint drug administration.
Consumer:Talk to your doctor before using probenecid together with glycerol phenylbutyrate. Probenecid may interfere with the excretion of metabolites of glycerol phenylbutyrate into the urine. Since blood and urine levels of these metabolites may be used to help guide glycerol phenylbutyrate treatment and determine effectiveness, your doctor will need to be aware of this interaction when making any dose adjustments. In addition, you may be at increased risk of developing nervous system side effects associated with one of the metabolites, including drowsiness, lightheadedness, fatigue, headache, taste disturbances, hearing problems, confusion, memory impairment, tremor, and seizures. You should contact your doctor immediately if you experience these or other unusual symptoms. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Professional:MONITOR CLOSELY: Probenecid may inhibit the renal excretion of metabolites of phenylbutyrate, including phenylacetate (PAA) and phenylacetylglutamine (PAGN). PAA is associated with neurotoxic effects such as somnolence, fatigue, lightheadedness, headache, dysgeusia, hypoacusis, disorientation, memory impairment, tremor, peripheral neuropathy, and seizures.
MANAGEMENT: Since plasma PAA and urinary PAGN levels may be used to help guide dosing of phenylbutyrate therapy, caution is advised if probenecid is coadministered. The potential for increased risk of PAA-induced neurotoxicity should also be considered.
