Professional:GENERALLY AVOID: Coadministration with potent inducers of CYP450 3A4 and/or P-glycoprotein (P-gp) may significantly decrease the plasma concentrations of copanlisib, which is a substrate of both the isoenzyme and efflux transporter. When a single 60 mg intravenous dose of copanlisib was administered to cancer patients in combination with the potent CYP450 3A4 and P-gp inducer, rifampin (600 mg once daily for 12 days), mean copanlisib peak plasma concentration (Cmax) and systemic exposure (AUC) decreased by 15% and 63%, respectively. Reduced efficacy of copanlisib may occur.
MANAGEMENT: Concomitant use of copanlisib with potent CYP450 3A4 inducers should generally be avoided.
