Consumer:

Using indinavir together with delavirdine may increase the effects of indinavir. Contact your doctor if you experience increased side effects or if your condition changes. If your doctor does prescribe these medications together, you may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Professional:

ADJUST DOSE: Coadministration with delavirdine may increase the plasma concentrations of indinavir. The mechanism is delavirdine inhibition of indinavir metabolism via CYP450 3A4. According to the delavirdine product labeling, coadministration of delavirdine (400 mg three times a day) and indinavir (600 mg three times a day) for 7 days increased the mean area under the plasma concentration-time curve (AUC) and trough plasma concentration (Cmin) of indinavir by 53% and 298%, respectively, compared to administration of indinavir 800 mg three times a day without delavirdine. When indinavir dosage was decreased to 400 mg three times a day and coadministered with delavirdine for 7 days, indinavir mean peak plasma concentration (Cmax) decreased by 36% while Cmin increased by 118% relative to administration of indinavir alone at 800 mg three times a day. Indinavir had no apparent effect on the pharmacokinetics of delavirdine based on comparison with historical data.

MANAGEMENT: Indinavir dosage should be reduced to 600 mg three times a day during coadministration with delavirdine.
