Darunavir Tablets and Incivek

In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. A doctor should be consulted to address the issue of joint drug administration.

Talk to your doctor before using darunavir together with telaprevir. Combining these medications may decrease the blood levels of both medications, which may reduce their effectiveness in treating hepatitis C and HIV infection. Your doctor may be able to prescribe alternatives that do not interact, or you may need a dose adjustment or more frequent monitoring by your doctor to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

GENERALLY AVOID: Coadministration of telaprevir and darunavir/ritonavir may decrease the plasma concentrations of both telaprevir and darunavir. The mechanism of interaction has not been described. In 14 study subjects, administration of telaprevir (750 mg every 8 hours for 10 days) in combination with darunavir/ritonavir (600 mg/100 mg twice daily for 20 days) reduced the telaprevir peak plasma concentration (Cmax), systemic exposure (AUC) and trough plasma concentration (Cmin) by an average of 36%, 35% and 32%, respectively, compared to administration alone. The Cmax, AUC and Cmin of darunavir were reduced by approximately 40% each during telaprevir coadministration.

MANAGEMENT: Given the risk of reduced viral susceptibility and resistance development associated with subtherapeutic drug levels, concomitant use of telaprevir and darunavir/ritonavir should generally be avoided.
