Darunavir Tablets and Indinavir

Using indinavir together with darunavir can increase the effects of both medications. Contact your doctor if you experience increased side effects which may include fever, sore throat, and headache with a severe blistering, peeling, and red skin rash. You may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

MONITOR: Coadministration of indinavir and darunavir/ritonavir may result in increased plasma concentrations of both indinavir and darunavir. The proposed mechanism is competitive inhibition of CYP450 3A4 metabolism. In nine study subjects, administration of indinavir (800 mg twice a day) with darunavir/ritonavir (400 mg/100 mg twice a day) increased the peak plasma concentration (Cmax), area under the concentration-time curve (AUC) and trough plasma concentration (Cmin) of indinavir by 8%, 23% and 125%, respectively, compared to administration of indinavir with ritonavir alone (800 mg/100 mg twice a day). Darunavir Cmax, AUC, and Cmin also increased by 11%, 24%, and 44%, respectively, compared to administration without indinavir.

MANAGEMENT: The appropriate dosage of indinavir when prescribed in combination with darunavir/ritonavir has not been established. Antiretroviral response should be monitored closely during treatment.
