Darunavir Tablets and Sertraline Oral Liquid

Using darunavir together with sertraline can decrease the effects of sertraline. Contact your doctor if your condition worsens. You may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

MONITOR: Coadministration with ritonavir- or cobicistat-boosted darunavir may decrease the plasma concentrations of sertraline. The mechanism of interaction has not been described. In 13 study subjects, administration of sertraline (50 mg once a day) in combination with darunavir/ritonavir (400 mg/100 mg twice a day) reduced the peak plasma concentration (Cmax), area under the concentration-time curve (AUC), and trough plasma concentration (Cmin) of sertraline by approximately one-half compared to administration without darunavir/ritonavir. Sertraline had no significant effect on the pharmacokinetics of darunavir.

MANAGEMENT: Patients treated concomitantly with ritonavir- or cobicistat-boosted darunavir may have diminished clinical response to sertraline. Pharmacologic response to sertraline should be monitored more closely whenever darunavir-ritonavir or darunavir-cobicistat is added to or withdrawn from therapy in patients stabilized on their existing antidepressant regimen, and the sertraline dosage adjusted as necessary.
