Consumer:Using zidovudine together with divalproex sodium may increase the effects of zidovudine. Contact your doctor if you experience unusual muscle pain and weakness, trouble breathing, fast or uneven heart rate, nausea, vomiting, stomach pain, and numbness or cold feeling in your arms or legs, diarrhea, loss of appetite, stomach pain, low fever, dark urine, clay-colored stools, or jaundice (yellowing of the skin or eyes). If your doctor does prescribe these medications together, you may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Professional:MONITOR: Valproate inhibits the glucuronidation of zidovudine. In six HIV-infected volunteers, concomitant use of zidovudine and valproic acid resulted in a twofold increase in the area under the plasma concentration-time curve of zidovudine. Elimination half-life was unchanged, suggesting an increase in oral bioavailability and a decrease in first-pass metabolism.
MANAGEMENT: Valproate may allow for reduced zidovudine dosage but not increased dosage intervals. Practitioners should be aware that the efficacy and toxicity profile of zidovudine may be altered in patients concomitantly receiving valproic acid or its derivatives.
