In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. A doctor should be consulted to address the issue of joint drug administration.

Erythromycin may significantly increase the blood levels of avanafil. This can increase the risk and/or severity of side effects such as low blood pressure, dizziness, lightheadedness, fainting, visual disturbances, ringing in the ears, vision or hearing loss, chest pain or tightness, irregular heartbeat, and priapism (prolonged and painful erection unrelated to sexual activity). Talk to your doctor if you have any questions or concerns. Your doctor may be able to prescribe alternatives that do not interact, or you may need a dose adjustment or more frequent monitoring to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

ADJUST DOSE: Coadministration with moderate inhibitors of CYP450 3A4 may increase the plasma concentrations of avanafil, which is primarily metabolized by the isoenzyme. In 15 healthy male volunteers, administration of a single 200 mg dose of avanafil following pretreatment with the moderate CYP450 3A4 inhibitor erythromycin (500 mg twice daily for 5 days) increased avanafil peak plasma concentration (Cmax) and systemic exposure (AUC) by approximately 2-fold and 3.6-fold, respectively, compared to administration of avanafil alone. Erythromycin also prolonged the half-life of avanafil to approximately 8 hours, from a normal of 5 hours.

MANAGEMENT: The dosage of avanafil should not exceed 50 mg once every 24 hours when used in combination with moderate CYP450 3A4 inhibitors.
