MONITOR: Coadministration of eslicarbazepine acetate with phenobarbital may decrease the systemic exposure to the active metabolite, eslicarbazepine. The proposed mechanism is phenobarbital induction of the glucuronidation of eslicarbazepine. According to the product labeling, eslicarbazepine systemic exposure (AUC) decreased by approximately 25% when eslicarbazepine acetate was administered with phenobarbital in healthy study subjects. Eslicarbazepine did not significantly affect the pharmacokinetics of phenobarbital.

MANAGEMENT: The potential for diminished therapeutic effects of eslicarbazepine should be considered when prescribed in combination with phenobarbital or primidone, a drug that is partially metabolized to phenobarbital. A dosage increase for eslicarbazepine acetate may be required based on efficacy and tolerability.
