Etravirine and Fosamprenavir

In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. A doctor should be consulted to address the issue of joint drug administration.

Ask your doctor before using fosamprenavir together with etravirine, this may increase the effects of fosamprenavir. Contact your doctor if you experience increased side effects or if your condition changes. If your doctor does prescribe these medications together, you may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

GENERALLY AVOID: Etravirine has been shown to significantly increase the plasma concentrations of amprenavir administered as fosamprenavir/ritonavir. The mechanism of interaction has not been described. In eight study subjects administered fosamprenavir/ritonavir (700 mg/100 mg) twice a day with etravirine, amprenavir peak plasma concentration (Cmax), systemic exposure (AUC) and trough plasma concentration (Cmin) increased by 62%, 69% and 77%, respectively. No data are available for fosamprenavir and etravirine without low-dose ritonavir as a pharmacokinetic booster.

MANAGEMENT: Appropriate dosages for the combination of etravirine and fosamprenavir/ritonavir have not been established. Due to a significant increase in amprenavir systemic exposure, fosamprenavir/ritonavir should not be prescribed in combination with etravirine. Unboosted fosamprenavir (i.e., without low-dose ritonavir) should also not be used with etravirine according to the product labeling for etravirine.
