Coadministration with dapagliflozin has been shown to slightly increase the plasma concentrations of simvastatin and its active metabolite, simvastatin acid, both of which are metabolized by CYP450 3A4. When a single 40 mg dose of simvastatin was given with a single 20 mg dose of dapagliflozin, simvastatin and simvastatin acid systemic exposure (AUC) increased by an average of 19% and 31%, respectively, compared to simvastatin administered alone. These changes are not considered clinically significant, and no dosage adjustment is recommended for simvastatin.
