In vitro data suggest that magnesium trisilicate may reduce the gastrointestinal absorption of warfarin. The mechanism of interaction is unknown. In an in vitro study involving absorption across a physiological membrane, cumulative absorption of warfarin was reduced 19.4% by magnesium trisilicate. Other antacids have not been reported to affect the pharmacokinetics of warfarin. In six healthy volunteers, aluminum-magnesium hydroxide given concurrently and for four additional doses every 2 hours had no effect on the plasma levels or hypoprothrombinemic effect of warfarin. Another study using aluminum hydroxide also found no evidence of an interaction with warfarin. No special precautions appear to be necessary when warfarin is prescribed in combination with antacids.
