Coadministration with cyclosporine, an inhibitor of CYP450 3A4 and organic anion transporting polypeptide (OATP), may modestly affect the pharmacokinetics of macitentan. In ten healthy male subjects pretreated with macitentan (30 mg initially, followed by 10 mg once daily) for 5 days, coadministration with cyclosporine (100 mg twice daily) on days 6 through 17 increased macitentan systemic exposure (AUC) by just 10% and trough plasma concentration (Cmin) by 38% compared to macitentan administered alone. These changes are not considered clinically relevant, and no dosage adjustments are recommended during coadministration.
