Gengraf (Cyclosporine Capsules, Modified) and Tracleer (Bosentan Tablets)
Determining the interaction of Gengraf (Cyclosporine Capsules, Modified) and Tracleer (Bosentan Tablets) and the possibility of their joint administration.
In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. A doctor should be consulted to address the issue of joint drug administration.
Consumer:Using cycloSPORINE and bosentan is not recommended. Taking these drugs together can cause symptoms of severe headache, nausea, decrease in blood pressure, or increase in the heart rate. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Professional:CONTRAINDICATED: Coadministration with cyclosporine may significantly increase the plasma concentrations of bosentan. The proposed mechanism is competitive inhibition of bosentan metabolism via CYP450 3A4 and/or inhibition of intestinal P-glycoprotein secretion by cyclosporine. According to the manufacturer, trough concentrations of bosentan were increased by approximately 30-fold during the first day of concomitant administration, and steady state concentrations were 3- to 4-fold higher than in the absence of cyclosporine. Conversely, the plasma concentrations of cyclosporine decreased by approximately 50%, probably due to induction of CYP450 3A4 by bosentan.
MANAGEMENT: Because elevated plasma levels of bosentan may be associated with increased risk of hepatotoxicity, the concomitant use of cyclosporine and bosentan is considered contraindicated.
