It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

GENERALLY AVOID: Coadministration with rifampin may significantly decrease the plasma concentrations of imatinib. The mechanism is rifampin induction of CYP450 3A4, the isoenzyme responsible for the metabolic clearance of imatinib. In 14 healthy volunteers, pretreatment with rifampin (600 mg orally once daily for 10 days) decreased the mean peak plasma concentration (Cmax) and area under the concentration-time curve (AUC) of imatinib (400 mg single oral dose) by 54% and 74%, respectively, compared to administration of imatinib alone. The clearance was increased 385% during rifampin coadministration. Although the pharmacokinetics of imatinib were subject to a high degree of intersubject variation, the interaction was reported in every subject.

MANAGEMENT: Due to the magnitude of the interaction that can potentially result in subtherapeutic plasma concentrations of imatinib, concomitant use with rifampin should be avoided if possible.
