Consumer:Antacids may interfere with the absorption of ledipasvir and reduce its effectiveness. Products with antacid effects typically contain aluminum hydroxide, calcium carbonate, magnesium hydroxide, magnesium carbonate, magnesium oxide, magaldrate, sodium bicarbonate, and/or sodium citrate. To prevent or minimize the interaction, antacid-containing preparations should be taken 4 hours before or 4 hours after the ledipasvir dose. Talk to your doctor or pharmacist if you have any questions or concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Professional:ADJUST DOSING INTERVAL: Concurrent administration of ledipasvir with drugs that elevate gastric pH such as antacids may reduce its gastrointestinal absorption. Ledipasvir exhibits pH-dependent solubility, with increased solubility at lower pH. Drugs that increase gastric pH are expected to decrease the plasma concentration of ledipasvir.
MANAGEMENT: If an antacid is required, the antacid should be taken 4 hours before or 4 hours after administration of ledipasvir-sofosbuvir. The same precaution may be applicable to other drugs with antacid effects.
