Consumer:

Using indinavir together with isavuconazonium can alter the blood levels of both medications. Specifically, indinavir levels may decrease, which may make the medication less effective in treating your HIV infection. At the same time, isavuconazonium levels may increase, and you may experience more frequent and/or severe side effects such as nausea, vomiting, diarrhea, and liver problems. Talk to you doctor if you have any questions or concerns. Your doctor may be able to prescribe alternatives that do not interact, or you may need a dose adjustment or more frequent monitoring by your doctor to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

Professional:

CONTRAINDICATED: Coadministration of isavuconazonium sulfate (prodrug of isavuconazole) and indinavir may increase the plasma concentrations of isavuconazole and decrease the plasma concentrations of indinavir. Both drugs are substrates as well as inhibitors of CYP450 3A4/5. The proposed mechanism for isavuconazole involves indinavir inhibition of CYP450 3A4/5-mediated metabolism of isavuconazole. The reduction in indinavir plasma concentrations occurs by an undetermined mechanism. Coadministration of isavuconazole with indinavir resulted in a decrease in indinavir AUC by 36% and Cmax by 52%.

MANAGEMENT: Concomitant use of isavuconazonium sulfate with potent CYP450 3A4 inhibitors, such as indinavir, is considered contraindicated.
