Coadministration with ritonavir may decrease the plasma concentrations of raltegravir. The proposed mechanism is ritonavir induction of the glucuronidation of raltegravir via UGT1A1. When a single 400 mg dose of raltegravir was administered to ten study subjects in combination with ritonavir 100 mg every 12 hours for 16 days, raltegravir peak plasma concentration (Cmax), systemic exposure (AUC) and trough plasma concentration (Cmin) decreased by 24%, 16% and 1%, respectively. The effects of ritonavir given at a dosage greater than 100 mg twice daily have not been evaluated.
