Professional:GENERALLY AVOID: Coadministration with potent inducers of CYP450 3A4 may decrease the plasma concentrations of ivosidenib, which is primarily metabolized by the isoenzyme. Based on physiologically-based pharmacokinetic modeling, coadministration of ivosidenib with the potent CYP450 3A4 inducer rifampin (600 mg once daily for 15 days) is predicted to decrease ivosidenib steady-state systemic exposure (AUC) by 33%. Reduced therapeutic efficacy may occur.
MANAGEMENT: Concomitant use of ivosidenib with potent CYP450 3A4 inducers should generally be avoided.
