Professional:CONTRAINDICATED: Coadministration with potent inducers of CYP450 3A4 may significantly decrease the plasma concentrations of lurasidone, which is primarily metabolized by the isoenzyme. In a pharmacokinetic study, administration of a single 40 mg dose of lurasidone with the potent CYP450 3A4 inducer rifampin (600 mg/day for 8 days) resulted in an 86% decrease in lurasidone peak plasma concentration (Cmax) and an 80% decrease in systemic exposure (AUC) compared to administration alone.
MANAGEMENT: Concomitant use of lurasidone with potent CYP450 3A4 inducers is considered contraindicated.
