Professional:Vemurafenib may decrease the plasma concentration of midazolam. The mechanism is related to induction of hepatic CYP450 3A4 enzymes responsible for the metabolism of midazolam. Administration of a single dose of midazolam after repeat dosing with vemurafenib for 15 days, resulted in a mean 39% decrease (maximum 80%) in midazolam plasma exposure. The clinical significance is unknown, but may depend on the dosage and duration of vemurafenib therapy.
