Professional:MONITOR: Theoretically, coadministration with acetaminophen may increase the plasma concentrations of busulfan. The proposed mechanism is decreased busulfan clearance via conjugation due to reduced levels of glutathione in blood and tissues caused by acetaminophen.
MANAGEMENT: Patients receiving busulfan therapy should be monitored more closely for toxicity (e.g., myelosuppression, seizures) if acetaminophen is administered concurrently. When busulfan is administered intravenously, acetaminophen should preferably be avoided within 72 hours prior to and during infusion of busulfan to minimize risk of an interaction.
