Coadministration with cyclosporine, a potent inhibitor of P-glycoprotein efflux transporter, does not appear to significantly alter the pharmacokinetics of sitagliptin, which is a substrate of P-glycoprotein. Following coadministration of a single 100 mg oral dose of sitagliptin and a single 600 mg oral dose of cyclosporine, the peak plasma concentration (Cmax) and area under the concentration-time curve (AUC) of sitagliptin were increased by 68% and 29%, respectively. These changes are not considered clinically significant. The renal clearance of sitagliptin was also not significantly altered in the presence of cyclosporine.
