Consumer:Using darunavir together with PARoxetine can decrease the effects of PARoxetine. Contact your doctor if your condition worsens. You may need a dose adjustment or special test to safely use both medications. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.
Professional:MONITOR: Coadministration with ritonavir- or cobicistat-boosted darunavir may decrease the plasma concentrations of paroxetine. The mechanism of interaction has not been described. In 16 study subjects, administration of paroxetine (20 mg once a day) in combination with darunavir/ritonavir (400 mg/100 mg twice a day) reduced the peak plasma concentration (Cmax), area under the concentration-time curve (AUC), and trough plasma concentration (Cmin) of paroxetine by approximately one-third compared to administration without darunavir/ritonavir. Paroxetine had no significant effect on the pharmacokinetics of darunavir.
MANAGEMENT: Patients treated concomitantly with ritonavir- or cobicistat-boosted darunavir may have diminished clinical response to paroxetine. Pharmacologic response to paroxetine should be monitored more closely whenever darunavir-ritonavir or darunavir-cobicistat is added to or withdrawn from therapy in patients stabilized on their existing antidepressant regimen, and the paroxetine dosage adjusted as necessary.
