Praziquantel and Rifadin

In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. A doctor should be consulted to address the issue of joint drug administration.

Using praziquantel together with rifAMPin is not recommended. Combining these medications may significantly decrease the blood levels of praziquantel and make it ineffective or less effective in treating your condition. Your doctor may be able to prescribe an alternative to praziquantel that you can take with rifAMPin, or you may need to discontinue rifAMPin for up to four weeks before you begin taking praziquantel. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

CONTRAINDICATED: Coadministration with potent CYP450 inducers such as rifampin may significantly decrease the plasma concentrations of praziquantel. In a crossover study with a 2-week washout period, plasma praziquantel concentrations were undetectable in 7 out of 10 subjects who ingested a single 40 mg/kg dose of praziquantel following pretreatment with oral rifampin 600 mg daily for 5 days. When the same dose of praziquantel was administered two weeks after discontinuation of rifampin, the mean praziquantel peak plasma concentration (Cmax) and systemic exposure (AUC) were 35% and 23% lower, respectively, than when praziquantel was given alone.

MANAGEMENT: The use of praziquantel in combination with potent CYP450 inducers such as rifampin is considered contraindicated because therapeutically effective blood levels of praziquantel may not be achieved. In patients receiving rifampin (e.g., as part of a combination regimen for the treatment of tuberculosis), alternative agents for schistosomiasis should be considered. However, if treatment with praziquantel is necessary, rifampin should be discontinued 4 weeks before administration of praziquantel. Treatment with rifampin can then be restarted one day after completion of praziquantel treatment.
