Rifampin and Vfend

In the database of official manuals used in the service creation an interaction registered by statistical results of studies was found, which can either lead to negative consequences for the patient health or strengthen a mutual positive effect. Using rifAMPin and voriconazole is not recommended. Call your doctor if you notice that voriconazole is not working or if your infection worsens. Talk to your doctor if you have any concerns. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

CONTRAINDICATED: Coadministration with rifampin may significantly reduce the plasma concentrations of voriconazole. The mechanism is rifampin induction of CYP450 isoenzymes that are responsible for the metabolic clearance of voriconazole. According to the manufacturer, rifampin (600 mg once daily) decreased the steady-state peak plasma concentration (Cmax) and area under the concentration-time curve (AUC) of voriconazole (200 mg every 12 hours for 7 days) by an average of 93% and 96%, respectively, in healthy subjects. Doubling the dose of voriconazole does not restore adequate systemic exposure to voriconazole during coadministration with rifampin.

MANAGEMENT: The use of voriconazole with rifampin is considered contraindicated.
