Secobarbital may reduce the blood levels of abemaciclib, which may make the medication less effective in some cases. Contact your doctor if your symptoms worsen or your condition changes. It is important to tell your doctor about all other medications you use, including vitamins and herbs. Do not stop using any medications without first talking to your doctor.

MONITOR: Coadministration with inducers of CYP450 3A4 may decrease the plasma concentrations of abemaciclib and its pharmacologically active metabolites, all of which are substrates of the isoenzyme. In healthy study subjects, administration of a single 200 mg dose of abemaciclib during 600 mg daily dosing of rifampin, a potent CYP450 3A4 inducer, decreased the relative potency-adjusted unbound systemic exposure (AUC) of abemaciclib plus its active metabolites by 67% relative to abemaciclib administered alone. Reduced efficacy of abemaciclib may occur. The interaction has not been studied with other, less potent inducers.

MANAGEMENT: The potential for diminished pharmacologic effects of abemaciclib should be considered during coadministration with CYP450 3A4 inducers. Alternative treatments may be required if an interaction is suspected.
