In 15 healthy male volunteers, administration of a single 50 mg dose of avanafil following pretreatment with the potent CYP450 3A4 inhibitor ketoconazole (400 mg once daily for 5 days) increased avanafil peak plasma concentration (Cmax) and systemic exposure (AUC) by approximately 3-fold and 13-fold, respectively, compared to administration of avanafil alone. Likewise, when a single 50 mg dose of avanafil was administered to 14 healthy male volunteers treated with ritonavir (600 mg twice daily for 5 days), avanafil Cmax increased by 2.4-fold and AUC increased by 13-fold. Both ketoconazole and ritonavir prolonged the half-life of avanafil to approximately 9 hours, from a normal of 5 hours.
