Coadministration with verapamil may increase the plasma concentrations of vemurafenib. The mechanism is inhibition of CYP450 3A4 and P-glycoprotein by verapamil. Vemurafenib has been shown in vitro to be a substrate of CYP450 3A4 and a weak substrate of P-glycoprotein. The clinical significance is unknown and clinical data are currently lacking.
