Methadose and Pegasys

Coadministration with peginterferon alfa-2a may increase the plasma concentrations of methadone. The mechanism of interaction is unknown. In a pharmacokinetic study of 24 chronic hepatitis C patients receiving stable methadone maintenance therapy (median dose 95 mg, range 30 mg to 150 mg), treatment with peginterferon alfa-2a (180 mcg/kg subcutaneously once weekly) for 4 weeks was associated with methadone levels that were 10% to 15% higher than at baseline. The clinical significance of this finding is unknown. It may be appropriate to monitor for signs and symptoms of increased narcotic effect during coadministration of peginterferon alfa-2a and methadone. Methadone did not significantly affect the pharmacokinetics of peginterferon alfa-2a as compared to a pharmacokinetic study of 6 chronic hepatitis C patients not receiving methadone.
