Consumer:Taking the riociguat and magnesium oxide doses too close together may interfere with the absorption of riociguat and reduce its effectiveness. Professional:ADJUST DOSING INTERVAL: Concurrent administration of riociguat with antacids may reduce its gastrointestinal absorption. Riociguat exhibits pH-dependent solubility, with increased solubility at lower pH. According to the product labeling, administration of riociguat with an antacid containing aluminum hydroxide and magnesium hydroxide resulted in decreases of riociguat peak plasma concentration (Cmax) and systemic exposure (AUC) by approximately 50% and 30%, respectively. The Cmax and AUC of the active metabolite, M1, which has 1/3 to 1/10 the pharmacologic activity of riociguat, were similarly reduced.
MANAGEMENT: Antacids should not be taken within 1 hour of riociguat administration.
