Coadministration with itraconazole has been shown to slightly increase the plasma concentrations of vandetanib, possibly by inhibiting the CYP450 3A4-mediated metabolism of vandetanib. In 16 healthy male subjects, coadministration of itraconazole (200 mg/day on days 1 to 24) and vandetanib (single 300 mg oral dose on day 4) resulted in a small but statistically significant 9% increase in vandetanib exposure (AUC) and an approximately 13% increase in half-life (from 209.2 hours to 235.5 hours) compared to vandetanib administered alone. These changes are not considered clinically relevant.
